Resumo:ABSTRACT:
Over the years, agriculture has applied various practices in soil management for agricultural production, considering that this practice has increased the levels of soil degradation and its quality, so that, consequently, interest in the identification of the microbiota in this environment. The main objective of this study is to isolate and identify microorganisms from three different agricultural management systems in Petrolândia - PE. Soil samples were taken from three irrigation projects located at the Itaparica dam in the municipality of Petrolândia-PE, identified as P1 - Apolônio Salles, P2 - Icó-Mandantes (Beginning) and P3- Icó-Mandantes (Final). Soil samples were collected in sterile plastic bags, packed in isothermal boxes and transported to the Laboratory of Molecular Biology of the Department of Biochemistry at UFPE. The isolation of bacteria was carried out by seeding different dilutions in methylene blue eosin agar and 5% bovine blood agar. The plates were placed in an oven at 37 ºC for 24 and 48 hours and after this period the readings were performed. Subsequently, the samples were identified using the Maldi-Tof technique (Bruker MALDI Biotyper). A total of 36 microorganisms were identified, resulting in 21 different species. The main genera found were Bacillus, Enterobacteria, Pseudomonas, Paenibacillus, so that the main species identified were: Bacillus cereus (28%), Enterobacter cloacae (11%), Stenotrophomonas maltophilia (8%), Bacillus megaterium (6%), and the others made up about 3% of each species, the main ones being: Pseudomonas nitroeducens, Clostridium difficile, Sthaphylococcus siniae, Klebsiella variicola, Brukhalderia gladioli, Pseudomonas putida, Empedobacter brevis, Paenibacillus agaridevorans. From the results obtained, it is concluded that the characterization of microorganisms in different soils under agricultural management in Petrolância-PE demonstrates a range of diversity of genera and species, of which, it is suggested the relevance of carrying out a monitoring of this microbiota, taking into account considering that these microorganisms are related to risks to the health of human beings and generate environmental impacts.
