INFLUENCE OF TRL-4 AND TRL-6 GENE SNP ON IL-6 LEVELS IN A POPULATION INFECTED WITH Plasmodium vivax IN A BRAZILIAN-FRENCH GUIANA BORDER POPULATION

Resumo:
Toll-like receptors (TLR) are one of the most interacting receptors during pathogen recognition. The TLR4 and TLR6 are present in the host cell, being expressed on the cell surface and interacting with extracellular ligands. They recognize pathogen-associated molecular patterns and are responsible for initiating the intracellular signaling cascade that stimulates the production of pro-inflammatory cytokines, among them IL-6, interferons, and chemokines. This study aimed to investigate the SNPs in the TLR4 and the TLR6 genes and its potential association with IL-6 serum levels in Plasmodium vivax infected patients from the Brazilian-French Guiana border. The sample group was composed of 144 non-malarial subjects and 76 Plasmodium vivax infected individuals with mild malaria. The rs4986790 A/G (C_11722238_20) and rs4986791 C/T (C_11722237_20) SNPs of TLR4 and the rs5743810 C/T (C_1180648_20) SNP for the TLR6 were genotyped using the qPCR technique (Applied Biosystems 7500 Fast Real-Time PCR System Life Technologies, MA, USA). The IL-6 Human Uncoated ELISA Kit was used to measure this cytokine level (Thermo Fisher Scientific, Waltham, MA, EUA). The statistical analyses were performed using the SPSS Software version 25.0. The observed genotypic frequencies were also compared with those expected according to the Hardy-Weinberg. The Association of polymorphisms with interleukin levels was evaluated by analysis of variance. The results obtained from the meta-analysis concluded that TLR-4, is associated with parasitemia, while TLR-6 is related to disease severity. No TLRs were associated with disease susceptibility. The results of genotyping between the investigated groups were not observed a significant association for the allele and genotypic frequencies of the studied SNPs between malaria-infected and uninfected individuals. An association between TLR4 (A299G) with parasitemia for the AA genotype of the malarial group. There was an increase for IL-6 in the TLR4 (A29T3911), TLR6 (S249P) SNPs for the malaria groups studied. The studied polymorphisms can modulate the parasitemia and the immune response of individuals infected by P. vivax in the Brazilian Amazon.
