Characterization of Microbial Communities Present in the Sediments of the Hydrographic Basin of the Yaque del Norte River in the Dominican Republic, Applying Metagenomic Techniques

Resumo:
The Yaque del Norte River in the Dominican Republic is the longest in the country. Its waters are used for agriculture, the industrial sector, domestic use and other activities. However, the quality of its waters is affected by different human activities. In this sense, the objective of this study was to analyze the microbial communities present in the sediments of the Yaqué del Norte river, taking into account the levels of anthropic contamination, which was determined according to the population present in each of the sampled points, to To achieve this, high-throughput metagenomics techniques (Ilumina) and other tools were used. The physicochemical results reveal higher levels of pH, EC, TDS, in the middle basin of the river (Otra Banda). At the taxonomic level, the bacterial domain represented 98% of all microorganisms, among the most abundant families are Rhodobacteraceae, Dechloromonas, Rhodocyclaceae, Conexibacteraceae, Bradyrhizobiaceae, Nitrospiras, Xanthomonadaceae, Pseudomonadaceae, Flavobacteriaceae, Streptomycetaceae. Among the most abundant genera are Burkholderia, Candidatus Accumulibacter, Dechloromonas, Mycobacterium, Pseudomonas, Anaerolinea, Anaeromyxobacter, Geobacter, Anaeromyxobacter, Desulfobulbus, Bradyrhizobium, Chitinophaga, Frankia, Nitrospira, Novosphingobium, Phaeodactylum, Rhodobacter, Streptomyces. Species richness was represented in the Otra Banda point, followed by Castañuela. The Simpson index showed a wide diversity of 0.9962 for two of the subpoints of the middle and lower zone of the basin. The Jaccard similarity indices, when comparing the points at a distance of 0.890 units, the rivers of the other Band and the Confluence showed a lot of similarity between their microbial communities. The Euclidean distance confirms the similarity between the rivers of the Otra Banda and Confluencia, with Castañuela being the least similarity of the rest of the points about abundance. It should be noted that this is the first metagenomic study carried out in the rivers of this country, and the results obtained will greatly benefit the health sector, the environment, and the scientific community in general.
