Resumo:Global warming and the discharge of pollutants from industrial activities represent serious threats to environmental sustainability, ecological integrity, and society. In this sense, research is being added in search of effective tools, such as enzymes, with the objective of mitigating pollution with nonspecific and toxic produts. One of the promising sources, still unexplored, for the biotechnological application of enzymes are the cyanobacteria and microalgae. Among the enzymes of industrial interest are the β-glycosidase that participate in the bioconversion of lignocellulosic material into simple sugars, making them an interesting option for more sustainable bioenergy production. In this project we aimed to produce and characterize the β-glycosidases from the cyanobacterium Microcystis aeruginosa CACIAM 03 in order to apply them in biotechnological processes. The enzyme production had been determined at 10 days from the incubation period. The β-glycosidase produced was characterized and properties of interest for industrial application were observed. The optimum activity of the crude enzyme extract was determined in pH 4.0 at 40 °C (28.51 U/mL) using β-pNPG as substrate. Furthermore, the enzyme was shown to have a broad spectrum of enzymatic activity between pH 3.5 and 6.5 losing its full activity at neutral pH. In addition, the enzyme had a range of activity between 35 and 50°C, maintaining 60% of its relative activity after 480 min of incubation, proving to be an enzyme with good thermostability in its activity range. The properties presented by β-glucosidase suggest that the enzyme is suitable for industrial applications, however, it is important to emphasize that further investigations and optimization of the production of β-glucosidases by cyanobacteria still need to be performed for large-scale application.
